Was this reprinted with the 1910 expansion? If I buy the 1910 expansion does this come with it?

No. 1910 includes the few additional tickets that were in Mystery Train, but does not include any of the special cards.

Half of it. Mystery Train had 4 new tickets and 5 "Role" cards which awarded points at the end of the game for accomplishing certain things. The new tickets are in 1910, but the Role cards are not.

In gmaeplay terms, do the Mystery Train routes do anything special or require anything special to achieve when playing 1910? I notice they are clearly labeled "Mystery Train" but there is no explanation for what this means in-game.

It's just a label to let you know those are the tickets that came from the Mystery Train expansion, they do not do anything special nor do they require anything special to achieve.

In the 1910 expansion, the route cards are all larger sized than the original game. They are marked MT only so that you can pull out all the expansion cards and play the original game with the base (yet larger sized) cards. That's really the only significance to them having Mystery Train on them.

AND

Since the 1910 expansion cards are larger, you can't really use the MT expansion cards with 1910, since the sizes are all wrong...
