When you're setting up the tiles for the Mansion, and one door doesn't connect to another door, is this door usable by the investigators to enter the next room?

No. There is a special tile to cover up the one door that doesn't connect so that you can avoid confusion.

Doors should always connect or else explicitly have a "blocked door" icon on the map to show it cannot be used.

Note that Green Eyed Boy has a couple of mistakes on it where the tiles are shown on the map in the wrong orientation, meaning the doors don't line up. Just rotate the tiles so that they do.

I don't have my copy of the game here but if I remember correctly there are doors that connects to an open side of a tile (and not another door), like a corridor then you can move through.
